Abstract

Angiogenesis appears to be an important event in the pathophysiology of Uterine Fibroids. Two angiogenic factors, fibroblast growth factor FGF 2 and VEGF, play a central role in the initiation of angiogenesis. We investigated whether the FGF2 754C/G and VEGF 460 C/T polymorphisms are associated with a risk of developing Uterine fibroids in Black Women Attending the Gynaecological Endoscopic Surgery and Human Reproductive Teaching Hospital- CHRACERH, Yaounde. A case-control study was performed on 60 consecutively collected blood samples, spotted on Whartman N° 3-filter paper from which DNA was extracted by the chelex-100 DNA extraction method. Genotyping of the FGF2 and VEGF SNPs was done using the Polymerase Chain Reaction and Restriction Fragment Length Polymorphism (PCR-RFLP). The Chi squared test (X2) was used to establish associations and a p-value of <0.05 was considered statistically significant. There was no difference in genotype distribution of the FGF2 745C/G polymorphism between uterine fibroid cases and controls (P>0.05), but the frequency of the C allele of FGF2 C745G polymorphism in the UF patients was lower than that of control women. The C allele showed to be a risk factor for the development of Uterine fibroids (OR=2.071, 95%CI=0.178-24.150). The Homozygous mutant genotype for the VEGF C460T showed a prevalence of 100% between the UF and control groups. In conclusion, our study demonstrated for the first time that the FGF2 745C/G VEGF 460C/T polymorphisms may be associated with a risk of developing Uterine fibroids in Cameroonian women. Carriers of the GG genotype of FGF2 C745G polymorphism could be protected against the development of Uterine fibroids.
